La Premier League 2009-10 sarà il 111° massimo campionato inglese di calcio, nonché diciottesima edizione della Premiership. La prima giornata si svolge il 15 agosto 2009 mentre l'ultima si disputerà il 9 maggio 2010.

Il campione in carica è il Manchester United. Le tre neopromosse sono Birmingham, Burnley e Wolverhampton, che rimpiazzano le retrocesse Newcastle, Middlesbrough e West Bromwich Albion.
